Yes-ish. The nearest is going to be Enterprise which is at 48th and 11th ave- about a 2 block walk. Hertz and Avis are about a 9 block walk south to 43rd.

Going to the port you can drop off your luggage at the porters, drive to return the car, and walk back to the pier.
