When you get of the ship the vans will be across the parking lot to take you to the car rental agencies. All the rental places are right there together, very quick trip. They have a shuttle that will pick you up from port and take you back to the office on Astronaut Boulevard. I'd recommend making reservations prior to the trip. In theory, Thrifty will work as well, but you can't book a one-way car rental (from Cape Canaveral to Orlando) on their website, you have to call the CC office directly, and there's a one-way drop off charge. Be aware that it's generally more expensive to rent from Cape Canaveral to MCO, versus from MCO to CC, so if you've already priced the airport-to-port leg, just be prepared to pay more on the way back.
