On a 5 day cruise, is there one formal night?    Is it night 2?   First Sea day night?

It depends on the order of your ports and sea days. It is never the first night nor the last night. So on a  day cruise it is day 2, 3, or 4. If one of those is a sea day, it is usually then.  If a port day, it is usually on the port day with the earliest back-on-board time.
