We will have $400 on board credit from cancelled Sensation cruise, can this credit be used in the casino (cash advance, etc...)?

 

And, will Carnival put as much of a hold on our credit card?

I would think so, but why not call Carnival and asked them directly.

Don't know why you would want to, there is a 3% charge to use your S&S card in the Casino. Also, a large chunk will disappear of the top for gratuities.
