I think you are tring to fit way to much into one day if you really want to see and enjoy especially St. Paul. Nice is a large city so you would have to decide what part you wanted to see.There is distance to consider as well with traffic.I am sure other's will ost with more info for you. If you want to go to St Paul de Vence (one of our favorite places in the South of France) then a rental car is a very good option. We would suggest picking up your car and driving directly to St Paul de Vence since it is your furthest drive from the ship. Enjoy that fabulous walled city and take the time to browse its galleries, shops, etc. Then you can drive back to Nice and consider your options with the remaining time. A big advantage to having your own car is the ability to be flexible and change your plans on the run. You may find that your plans are a bit too ambitious for the allotted time, and might want to make some plan changes as your day progresses. Personally, we think you are trying to do too much and will find that you will have to rush through everything (not fun) in order to meet all your objectives. I might suggest going straight to the Villa Ephrussi deRothchild after St Paul de Vence, and then if you still have some extra time you might be able to make a quick stop at Eze Village before returning to the port. You can certainly drive through the coastal part of Nice on your way to the Villa Rothchild, but might not want to spend the time stopping in this large city. Car in Monaco, St Paul de Vence, Villa Rothchild and possibly Eze on return. Do you use any particular car company? Do you get better rates from one than another?
