There is detailed information available on the park web site; 90 minutes is the posted time for restroom stops. You stop for about 15 minutes. At Eielson you stop for 30-45 minutes depending on how many wildlife stops you made on the way.

The restrooms aren't flush toilets but they aren't nasty blue porta potties. There are photos of the restrooms and rest areas in the following trip report if you look down the page.

The stops are about every 90 minutes, same as the tour buses. MAJOR reason, my minimum distance is Eielson. Nice building with informative exhibits and running water and flush toilets. ALL toilets before this point are chemical.
