We are scheduled to go on the Pride in October, but my wife just found out that she can't get the time off of work. Is there a penalty to change her reservation over to my girlfriend's?

I don't know how Carnival works but on other cruise lines, you can change the second passenger as long as the primary passenger stays on the booking without penalty assuming it is not within final payment.

Your best bet would be to call your TA or Carnival directly.

It depends on what rate you booked under. Some rates will be no penalty and others there could be a penalty.
