You can get there by HOHO bus or metro but, easiest is to take a taxi which cost 10 Euro from the cruise port last year.  It is an excellent tour.

I highly recommend the tour. Be prepared to spend some time (and money!) in the huge gift shop, too.  We went here twice in the three days we were in Barcelona.
