In-laws can't go, can my friend and wife take theirbookoing? Cost? or rebook?

As I understand it (I'm sure someone will be along to correct me)... one name on the reservation needs to remain, the other names can be changed with no hassle. What I can't tell from your post is if you're referring to two people out of four travelling on the same reservation number, or two people with their own reservation number.

 

If it's the former, should be no problem to change the names. If it's the latter, I'm guessing a cancel and rebook. If that's the case, I would arrange a three way call between the people cancelling, the people booking, and the travel agent (or RCI if you booked directly) to make sure the rooms transfer (assuming the new folks want the same room the old folks were in).

My family booked, piggybacked on our on-board booking...

 

They decided they can't go. So, it's before final payment.

 

Just wondering if my BF and his DH take over this reservation which is in Jan 2017?

Sounds like the answer is no as 1 persons name from the original reservation must remain on the booking.
