To address the "must sees":  there are so many in Rome that you will have to do some research and decide what's important.

 

Giving the crowds of recent years, it is unlikely that you will be able to see IN INSIDE VISITS what are arguably the two major "must sees" -- the Vatican (including St. Peter's basilica and the Vatican museums) and the Colosseum. You could like do one of them with an inside visit and the other with a quick stop and view of the exterior only. 

 

I suggest you get hold of a guidebook or two about Rome, and/or watch some YouTube videos, documentaries about Rome's history and sites. Then if you are interested in a private tour, you will at least have some opinions about what you want to see and do.

The tour companies do a great job getting you to the highlights and can skip the line ups. With one day this is really the way to go. We did Share an Shore Excursion, years ago, Rome in Limo also offers shared options for cruise passengers.  Otherwise I'd say book ship transport into Rome and plan to map out a walking tour where you can see one highlight, maybe the Colosseum and then spend time walking around one or two adjacent parts of the city, going into churches, sitting in a famous Piazza and maybe having a nice lunch.
